產品描述：β-分泌酶（β-Secretase , EC 3.4.23.46），也稱為BACE1（APP切割酶的β-位點）或 memapsin-2（膜相關的天冬氨酸蛋白酶），是天冬氨酸 – 蛋白酶。與γ-分泌酶 ( γ-secretase) 一起，BACE1切割APP（Membrane-anchored amyloid precursor protein-膜錨定的澱粉樣蛋白前體蛋白）並產生可溶性澱粉樣蛋白-β（soluble amyloid-β (Aβ)）。 Aβ的累積是阿爾茨海默病 ( Alzheimer’s disease) 發病的關鍵事件，因此監測和靶向BACE1活性對於研究和開發可以緩解/治愈阿爾茨海默病的治療策略是重要的。最近的研究表明，用合成化合物抑制BACE1活性有助於減緩或阻止阿爾茨海默病的症狀。 BioVision的BACE1檢測試劑盒II提供了一種快速簡便的方法，可用於測量各種樣品中的BACE1活性。在該試劑盒中，分泌酶特異性肽與兩種報告分子EDANS和DABCYL綴合。在未切割的形式中，來自EDANS的螢光發射通過DABCYL部分的物理接近而猝滅。通過BACE1切割肽分離EDANS和DABCYL基團，允許釋放強螢光信號（Ex / Em = 345 / 500nm）。樣品中分泌酶活性的水平與螢光強度水平成比例。該測定簡單，靈敏，高通量適應性，並且可以檢測每個樣品低至0.2μU的BACE1活性。
