線粒體跨膜電位的破壞是誘導凋亡後發生的最早的細胞內事件之一。 MitoCapture™細胞凋亡檢測試劑盒提供了一種簡單的基於螢光的方法，通過檢測線粒體跨膜電位的變化來區分健康細胞和凋亡細胞。該試劑盒使用MitoCapture™，一種陽離子染料，其在健康和凋亡細胞中發螢光不同。在健康細胞中，MitoCapture在線粒體中積累和聚集，產生明亮的紅色螢光。在凋亡細胞中，由於線粒體跨膜電位的改變，MitoCapture不能在線粒體中聚集，因此它保留在其單體形式的細胞質中，呈綠色螢光。螢光信號可以通過使用帶通濾光片（檢測FITC和羅丹明）的螢光顯微鏡檢測，或通過流式細胞術使用FITC通道進行綠色單體（Ex / Em = 488/530 + 30nm）和（可選）PI紅色聚集體的通道（Em = 488/590 + 42nm）。
