CheKine™ Micro Urea Nitrogen Content Detection Kit

尿素(Urea) 是生物體內含氮化合物(nitrogenous compound )降解的最終產物，在脲酶(urease)的催化下轉化為氨。血中尿素氮（Blood urea nitrogen, BUN）是腎功能的主要指標之一。在樣品中，尿素氮在含有氯化鐵和磷酸鹽以及二乙醯單肟和硫脲的溶液中發生反應，加熱後生成紅色二嗪化合物。該化合物的顏色強度與存在的尿素氮濃度成正比。尿素氮水平的測定是使用二乙醯單肟法(diacetyl monoxime method)完成的，該方法依賴反應混合物的比色分析來定量血清或尿液樣本中的尿素氮含量( urea nitrogen content)。

Urea is the end product of nitrogenous compound degradation in living organisms, undergoing conversion to ammonia under the catalysis of urease. Blood urea nitrogen (BUN) is one of the principal indicators of renal function. In a sample, urea nitrogen reacts in a solution containing ferric chloride and phosphate, along with diacetyl monoxime and thiourea, upon heating to produce a red diazine compound. The intensity of the color of this compound is directly proportional to the concentration of urea nitrogen present. The determination of urea nitrogen levels is accomplished using the diacetyl monoxime method, which relies on colorimetric analysis of the reaction mixture to quantify urea nitrogen content in serum or urine specimens.

 

樣本製備:

1. 組織樣本：秤取 0.1 g 組織樣本，加入 1 mL 去離子水，冰上勻漿。 10,000 g 離心 10分鐘在 4℃。取上清液進行測定，置於冰上待測。
2.細胞：收集5×106個細胞至離心管中，用冷PBS洗滌細胞，離心後棄上清；添加 1mL 去離子水超音波破碎細胞或細菌 3 min（功率 300 W，超音波 3 s，間隔 7 s，總時間 3分鐘）。 4℃ 10,000 g 離心 10 分鐘。取上清液進行測定，置於冰上待測。
3.血清（血漿）等液體樣本：直接測定。如果液體混濁，則用上清液測定離心

 

注意：如果需要測定樣本的蛋白質濃度，建議使用 Abbkine貨號KTD3002 蛋白質定量試劑盒（Bradford Assay）測量樣本的蛋白質濃度。

測定程序:
1.將酶標儀或可見分光光度計預熱30分鐘以上，調整波長至540 nm，可見光用去離子水將分光光度計調零。
2、操作表（以下操作均在1.5 mL EP管中進行）：

充分混合，然後在沸水浴中孵育 10 分鐘。冷卻後，吸移 200 μL 至96 孔板。測量 540 nm 處的吸光度
