Background	One of the most easily measured features of apoptotic cells is the break-up of the genomic DNA by cellular nucleases. Terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) is a method for detecting DNA fragmentation by labeling the 3′-hydroxyl termini in the double-strand DNA breaks generated during apoptosis. The TUNEL assay relies on the presence of nicks in the DNA which can be identified by TdT, an enzyme that catalyzes the addition of dUTPs that are labeled with fluorescein.

A.樣品製備

—-用於貼壁細胞(adherent cells)（螢光顯微鏡分析）

1. 96-well microplate culture 生長至少24h。通過誘導細胞凋亡所需的方法。同時培養無誘導的對照組。
2. 移去培養基，並在PBS中用4％多聚甲醛固定細胞室內溫度30分鐘。
3. 除去固定液並用PBS清洗3次（每次5分鐘）。
4. 固定後，添加通透劑（PBS中0.2％Triton X-100，未提供），並將板在室溫下孵育30分鐘。用PBS洗滌細胞2-3次。繼續執行步驟B 1。

最佳化: 製備TUNEL反應的陽性對照( positive control): 室溫下將含有10 U / mL DNAase I的細胞, 在室溫下放置10分鐘，然後再進行TUNEL操作反應（步驟B 1）。

—-用於非貼壁細胞 （通過流式細胞儀分析）

1. 培養細胞至最佳密度（約1-2 x 10⁶細胞/ mL）。通過誘導細胞凋亡所需的方法。同時培養無誘導的對照培養物。
2. 通過300g離心收集1-5 x 10⁶個細胞。用0.5 mL PBS洗滌兩次。
3. 加入1 mL 4％多聚甲醛（在PBS中，pH 7.4）並在冰上孵育30分鐘。
4. 以300g離心細胞。除去上清液，重懸於1 mL PBS中。重複此洗滌兩次。
5. 在室溫下將細胞在0.3％Triton-X 100中重懸5分鐘以通透（或者，將細胞重懸於100μg/ mL蛋白酶K中5分鐘以使其通透）。
6. 以300g離心細胞。除去上清液，重懸於1 mL PBS中。

重複此洗滌兩次，然後繼續步驟B 2。
—–用於石蠟包埋的組織（通過螢光顯微鏡分析）

1. 通過將兩次在二甲苯中浸泡10-20分鐘來對組織進行脫蠟。
2. 通過以下洗滌（給定順序）為組織補水：兩次洗滌5分鐘
   c. 每次在100％乙醇中洗滌，然後依次在95％，70％和50％乙醇中每次洗滌3分鐘。

B. Tunel分析

螢光顯微鏡分析

1. 在使用前根據樣品數量準備TdT標記反應緩衝液被檢測：向每個樣品中加入50 µL反應混合物（來自步驟A 1、3和4），並孵育在37 oC下放置60分鐘。
2. 將樣品在PBS中洗滌3次，每次5分鐘。 通過在1 µg / mL碘化丙啶或0.05 µg / mL DAPI中孵育而對樣品進行複染PBS 10分鐘

注意：根據組織的不同，可能必須調整復染劑的濃度。碘化丙錠或DAPI過度染色可能會導致觀察螢光素標記。

1. 將樣品在PBS中洗滌3次，每次5分鐘。

流式細胞術分析
