The kit provides a simple method for detecting TC concentration in a variety of biological samples such as serum, plasma, tissues, cells, and bacteria. In the assay, esterase catalyzes the hydrolysis of cholesterol esters to produce free cholesterol (FC) and free fatty acids (FFA). Then Cholesterol oxidase catalyzes free cholesterol (FC) to produce △4-cholestenone and H2O2. Further, H2O2, 4-aminoantipyrine and phenol can be catalyzed by peroxidase to form red quinone compounds which has a characteristic absorption peak at 500 nm. The Total Cholesterol (TC) present in the sample is proportional to the signal obtained.Total cholesterol (TC) includes free cholesterol and cholesterol esters. Tissue total cholesterol (TC) refers to the sum of cholesterol contained in all lipoproteins in the tissue.

(文獻資料) Serum triglycerides (TG), total cholesterol (TC), high density lipoproteins (HDL), and low density lipoproteins (LDL) were estimated in the five groups that each had eight rats in it. The diabetic rats displayed significantly high levels of serum triglycerides, total cholesterol and LDL, as well as a low level of HDL, as shown in Table 4. The treatment of C. papaya alleviated dyslipidemia in the Diab + C. papaya group, which was similar to the metformin treatment. The control rats treated with C. papaya did not show any changes.

KTB2220試劑盒可以用於檢測雞蛋樣本，蛋白樣本可直接測定; 蛋黃樣本按操作手冊中，組織樣本處理方法製備即可。(如下所示) 樣品製備注意：建議使用新鮮樣品，如不立即檢測，樣品可在-80℃保存一個月。(一)動物組織：稱取約0.1g組織，加入1mL無水乙醇。在冰上均質化。 4℃ 8,000g 離心 10 分鐘。使用上清液進行測定，並將其置於冰上進行測試。(二) 細胞或細菌：收集 5×10^6.將細胞或細菌放入離心管中，離心後棄上清；加入1mL無水乙醇在冰浴中超聲破碎細胞或細菌5分鐘（功率20%或200W，超聲3s，間隔7s，重複30次）。 4℃ 8,000 g 離心 10 分鐘。使用上清液進行測定，並將其置於冰上進行測試。(三) 血清（血漿）樣品：直接檢測。注：如果以蛋白濃度計算含量，最好用Protein Quantification Kit (BCA Assay), Cat #: KTD3001對總蛋白進行定量。
