葡萄糖氧化酶 (glucose oxidase enzyme , GOx) (EC 1.1.3.4)是一種氧化還原酶，可催化葡萄糖有氧氧化成過氧化氫(H ₂ O ₂ )和葡萄糖酸。葡萄糖氧化酶 (glucose oxidase enzyme , GOx) (EC 1.1.3.4) 是一種二聚酶(dimeric enzyme)，可催化 β-D-葡萄糖(beta-D-glucose)氧化成過氧化氫(hydrogen peroxide)和 D-葡萄糖酸-1,5-內酯(D-glucono-1,5-lactone)，後者被水解成葡萄糖酸(gluconic acid)。 這種酶是由某些種類的昆蟲(insects)、真菌(fungi)和細菌(bacteria)產生的。葡萄糖氧化酶(Glucose oxidase)廣泛用於測定體液中的葡萄糖以及去除飲料、食品和其他農產品中殘留的葡萄糖和氧氣(oxygen)。 微生物葡萄糖氧化酶因其在化學、製藥、食品、飲料、臨床化學、生物技術等行業的廣泛應用而備受關注。近年來，葡萄糖氧化酶在生物感測器中的新應用增加了需求。此外，葡萄糖氧化酶(Glucose oxidase)通常用於生物傳感器(biosensors)以檢測葡萄糖。廣泛應用於工業原料生產、生物感測器和癌症治療等領域。

CheKine™ 葡萄糖氧化酶活性測定試劑盒(CheKine™ Glucose Oxidase Activity Assay Kit, KT1310)提供了一種簡單易行的比色測定法，用於檢測生物樣品中的葡萄糖氧化酶活性(Glucose Oxidase Activity)，例如血清或血漿、細胞、尿液、其他體液、食物、生長培養基中的葡萄糖氧化酶活性。 在該測定中，葡萄糖氧化酶 (glucose oxidase enzyme , GOx) 活性通過偶聯酶測定確定，其中葡萄糖氧化酶 (glucose oxidase enzyme , GOx) 氧化 D-葡萄糖(D-glucose)，導致產生過氧化氫 (hydrogen peroxide , H2O2)，該過氧化氫 (hydrogen peroxide , H2O2) 在酸性條件下與色原反應，產生可在 OD 580 下測量的產物。 因此，樣品中存在的葡萄糖氧化酶活性與獲得的信號成正比。偵測範圍: 0.05 – 2 U/L, 最低靈敏度0.05 U/L。可應用於以下樣品來源: serum or plasma, cells, urine, other body fluid, food, growth medium。

1. Reaction.

將 50 μL 稀釋的標準品和樣品轉移到適當標記的管中。 加入 50 μL 0.2 M 葡萄糖。 緊緊關閉管子並混合。 將管子放在管架上，在 37°C 下孵育 20 分鐘。

Transfer 50 µL diluted standards and samples to appropriately labeled tubes. Transfer 50 µL 0.2 M Glucose to each tube. Close the tubes tightly and mix. Place the tubes in a tube holder and incubate for 20 min at 37°C.

2. 將 60 µL 轉移到底部透明的 96 孔板中。 每Well快速添加 40 µL  Chromogen 。 輕敲盤子以簡單徹底地混合。 立即讀取 580nm (OD0) 處的光密度。 37°C 避光孵育 10 分鐘。 再次讀取 580nm 處的光密度 (OD10) 化驗程序

Transfer 60 µL into a clear bottom 96-well plate. Add 40 µL of Chromogen per well quickly. Tap plate to mix briefly and thoroughly. Immediately read optical density at 580nm (OD0). Incubate for 10 min at 37°C in the dark. Read optical density at 580nm again (OD10)
