穀胱甘肽 (Glutathione) 研究的意義
穀胱甘肽是含有谷氨酸 (glutamate),半胱氨酸(cysteine)和甘氨酸(glycine)的三肽,其中含有巰基(sulfhydryl groups)。它廣泛存在於動植物中。它具有抗氧化劑(antioxidant)和綜合解毒(detoxification)作用。其相關的輔酶(coenzyme)是NADPH。人體新陳代謝產生的自由基(free radicals)過多會破壞生物膜(biofilm),侵入生命大分子,加速人體衰老(aging),並誘發腫瘤(tumors)或動脈粥樣硬化(atherosclerosis)的產生。穀胱甘肽具有增強生物體的抗氧化功能,促進生長以及增強生物體對人體不利環境的影響的功能。它的主要生理作用是清除體內的自由基。它也是一種很好的解毒劑,可以減弱有毒物質的毒性並從體內清除有毒物質。穀胱甘肽還促進生長並提高人體免疫力。穀胱甘肽(Glutathione)不僅可以用作藥物,還可以用作功能性食品的基礎材料,並且廣泛用於功能性食品中,例如抗衰老(anti-aging) ,增強免疫力(enhancing immunity)和抗腫瘤作用(anti-tumor)。
穀胱甘肽有兩種形式:還原型(GSH)和氧化型(GSSG)。氧化穀胱甘肽(Glutathione oxidized),也稱為二硫代穀胱甘肽(Dithioglutathione),其是通過兩個還原的穀胱甘肽分子的氧化形成的。穀胱甘肽還原酶將GSSG還原為GSH。還原型穀胱甘肽存在於大量生物中,並起主要作用。在紅血球細胞中,還原型穀胱甘肽(GSH)是保持血紅蛋白處於還原狀態並保護細胞免受氧化損傷的關鍵。 GSH是細胞中最重要的抗氧化劑巰基化合物,在抗氧化性,蛋白質巰基保護和氨基酸跨膜運輸中起重要作用。還原型和氧化型穀胱甘肽之間的比例(GSH / GSSG)是細胞氧化還原狀態的主要指標。因此,測量細胞中GSH和GSSG的含量以及GSH / GSSG的比值可以反映細胞的氧化還原狀態,但是其他具有過氧化活性的酶卻沒有利用這些底物。穀胱甘肽(Glutathione)檢測 (貨號KTB1600) 套組: 原理DTNB與還原型穀胱甘肽反應形成黃色產物。在412 nm處測得的光密度可以直接反映樣品中的穀胱甘肽濃度。
