POD過氧化酶(Peroxidase)廣泛存在於植物體內的不同組織,屬活性較高的誘導酶,能夠反應植物的代謝狀況和對環境的適應性。 POD 過氧化酶(Peroxidase)具有雙重的作用特性,在逆境或衰老初期的表現為清除 H2O2 ,達到保護細胞的功能。但在逆境或衰老後期,則參與活性氧的生成、葉綠素的降解,引發膜脂過氧化作用,加速細胞的衰老死亡,成為植物的衰老指標。植物體清除活性氧的保護系統又稱為抗氧化系統, 包括抗氧化酵素及非酵素型抗氧化 劑 (表 2 ),以應付具擴散性、水溶性及可與各種生物分子反應之活性氧化物,避免發生使活性物種毒性加強的連鎖反應。(出自農業部植物對環境逆境之調控與應用100年9月(第231期)。
過氧化酶(Peroxidases , EC編號1.11.1.x)是一大類酶,通常催化以下形式的反應:ROOR’+電子給體(2 e-)+ 2H +→ROH + R’OH。對於這些酶中的許多酶,最佳底物是過氧化氫(hydrogen peroxide),但其他酶則對有機氫過氧化物(organic hydroperoxides)(例如脂質過氧化物, lipid peroxides)更具活性。過氧化物酶(Peroxidases)可在其活性位點(active sites)包含血紅素輔因子(heme cofactor),或在氧化還原活性的半胱氨酸(redox-active cysteine)或硒代半胱氨酸殘基(selenocysteine residues)上具有替代性。過氧化物酶(Peroxidase )偶聯物被廣泛用作ELISA,免疫組織化學技術以及Northern,Southern和Western印跡分析的輔助檢測試劑。過氧化物酶,POD(EC 1.11.1.7)廣泛存在於動物、植物、微生物和培養細胞中。能催化過氧化氫(hydrogen peroxide)、酚類( phenols )和胺類化合物(amine compounds)的氧化,並消除過氧化氫(hydrogen peroxide)、酚類(phenols )和胺類(amines)。毒性的雙重作用。CheKine™ Micro過氧化酶分析試劑盒,該試劑盒提供了一種檢測植物組織(plant tissues)、細菌(bacteria)、真菌(fungus )或細胞等多種生物樣本中 POD 活性的簡單方法。在測定中,POD透過H2O2催化特定底物的氧化,產生在460nm處具有特徵光吸收的有色物質。
