彗星分析之應用 彗星分析又稱單細胞凝膠電泳分析，為檢測細胞內DNA損傷提供簡便有效的方法。自成功地用於檢測DNA單鏈斷裂以來，經不斷地改進，已成為一種快 速、靈敏和簡便地檢測DNA損傷地方法，廣泛地應用於DNA放射損傷、DNA剪切損傷、DNA交聯的檢測、藥物的遺傳毒性評價和細胞凋亡鑒定等工作中。普通的彗星分析及螢光染色產生的資料結果需要螢光顯微鏡進行觀察，並且不易保存，如果能獲得永久的彗星分析圖像結果，並可在普通顯微鏡下檢測則可以提高彗星分析的便捷性。

Comet Assay Kit (3-Well Slides)
DNA損傷介紹: 正常代謝活動和環境因素引起的DNA損傷，引發DNA損傷反應激活DNA修復途徑。 在DNA不可逆之情況下，誘導細胞程序凋亡。 正常DNA結構如DNA斷裂，DNA adducts 或交聯，需要DNA過程來修復。 DNA修復需要100多個基因。
彗星分析的檢測原理: 是基於變性的原理，待測細胞被裂解處理以使DNA解旋和變性，之後細胞核進行電泳，使得細胞在彗星玻片上平鋪的低熔點瓊脂糖上移動，之後使用與DNA相互作用的螢光染料或銀染試劑進行 染色使結果可視。形成的DNA彗星尾形狀和遷移模式，包括鹼性彗星尾長度，尾部DNA百分比以及尾矩的計算等資料可使用圖像分析軟體進行，最終用於評估細胞中DNA的損傷。

產品介紹
Comet 檢測試劑盒（3 孔載玻片）是一種單細胞凝膠電泳檢測 (single cell gel electrophoresis assay, SCGE)，可快速靈敏地檢測細胞中的 DNA 損傷(DNA damage )。其原理是器官或組織遭受，如輻射( radiation)、重金屬等(heavy metals)損傷後，細胞內的單鍊或雙鏈DNA(double strand DNA)斷裂，將細胞與熔化的瓊脂糖(melted agarose)混合後加入彗星載玻片(CometSlide)上，然後細胞膜( cell membrane )被裂解液( lysis solution)破壞，DNA被鹼性溶液(alkaline solution)解開。最後將樣品進行電泳(electrophoresed)，在電場作用(electric field)下，正常細胞的大分子DNA(macromolecular DNA)遷移距離較短，完整的DNA留在細胞核(nucleus)範圍內，DNA碎片游離出細胞核(nucleus)，PI染色染料，在螢光顯微鏡下觀察，完整的DNA形成圓形或微帶尾的圖案； DNA 碎片片段形成彗星狀圖案(comet-like pattern)。

彗星分析操作流程：
注意事項：試劑新鮮配製；對於含有血紅素的樣品，例如血細胞和組織，DMSO 可以以 10% 的終濃度添加。使用前將 1×Lysis Buffer 在 4℃ 冷卻至少 20 分鐘。 1×LysisBuffer在室溫下呈混濁狀，4℃時會變澄清，pH值保持在10左右。

彗星分析資料分析 (Data Analysis The DNA damage)
數據分析 DNA 損傷是通過測量細胞核遺傳物質（“彗星頭(comet head)”）和由此產生的“尾巴”之間的位移來量化的。 Tail Moment 和 Tail DNA% 是分析 Comet 檢測結果的兩個最常用參數。每個樣品至少應分析 50-100 個細胞。考慮到遺傳物質的遷移以及尾部 DNA 的相對量，尾矩(Tail Moment )被認為是誘導 DNA 損傷的適當指標。
Data Analysis The DNA damage is quantified by measuring the displacement between the genetic material of the nucleus (‘comet head’) andtheresulting ‘tail’. Tail Moment and Tail DNA% are the two most common parameters to analyze Comet assay results. At least 50-100cells should be analyzed per sample. The Tail Moment has been suggested to be an appropriate index of induced DNAdamageinconsidering both the migration of the genetic material as well as the relative amount of DNA in the tail.
Tail DNA%=Tail DNA Intensity/Cell DNA Intensity×100
Tail Moment can be measured using one of the following methods:
(a) Olive Tail Moment=Tail DNA%×Tail Moment Length (measured from the center of the head to the center of the tail, seeFigure1)
(b) Extent Tail Moment=Tail DNA%×Tail Length( see Figure 1) Comet Assay results analysis software is available through Opencomet, CASPlab and Comet Assay IV (Perceptive Instruments),among others.
