碳水化合物(Carbohydrate)是動植物的重要成分之一，也是新陳代謝(metabolism)的主要原料和儲存物質。總碳水化合物(Total carbohydrates)主要是指在測定條件下可水解成還原性單醣(reducing monosaccharides)的還原性葡萄糖(reducing glucose)、果糖(fructose)、麥芽糖(maltose)、乳糖(lactose)、蔗糖(sucrose)和麥芽糖以及可能部分水解的澱粉。CheKine™ 總碳水化合物檢測試劑盒（Total Carbohydrate Assay Kit, 比色法）提供了一種簡單、方便、快速的比色總碳水化合物(total carbohydrate)檢測方法。試劑盒原理是將總碳水化合物(total carbohydrate)水解成還原糖(reducing sugarsthe)，還原糖(reducing sugarsthe)與DNS試劑在鹼性條件(alkaline conditions)下加熱還原DNS生成氨基化合物(amino compounds)，在鹼性溶液(alkaline solution)中呈紅棕色，在540 nm處有特徵吸收峰.在一定濃度範圍內，還原糖含量與540 nm處的吸光度呈線性關係。根據標準曲線，可以計算出樣品中的總碳水化合物(total carbohydrate)含量。

樣品製備(Sample Preparation)

1.植物動物組織樣品：
稱取0.1g組織，加1mL鹽酸、1.5mL去離子水，均質，沸水浴30min。然後加入1mL氫氧化鈉，混勻，用去離子水稀釋至10mL。 8,000g在25℃下離心10分鐘。上清液用於測定。
2.血清或血漿樣品：取0.1mL血漿（血清），加0.1mL鹽酸、0.15mL去離子水，勻漿，沸水浴30min。然後加入0.1mL氫氧化鈉，混勻，用蒸餾水稀釋至1mL。 25°C下8,000g離心10分鐘。使用上清液進行測定。注：如果按蛋白質濃度計算含量，最好用ProteinQuantificationKit(BCAAssay)，Cat#：KTD3001來定量總蛋白。
