脂肪酶（Lipase, LPS）又稱為甘油酯水解酶( glycerol ester hydrolase)，能催化三酸甘油酯(triglycerides)水解，生成遊離脂肪酸(free fatty acids) 和甘油(glycerol)（或稱二甘油酯和單甘油酯）。 LPS廣泛分佈於各種生物體中。在胰臟炎(pancreatitis) 和胰臟癌(pancreatic cancer)中，血清中的脂肪酶（Lipase, LPS）水平通常會升高。 CheKine™ 微量酸性磷酸酶（ACP）活性測定試劑盒(CheKine™ Micro Acid Phosphatase (ACP) Activity Assay Kit) 提供了一種簡單、方便、快速的檢測脂肪酶（Lipase, LPS）活性的方法，適用於動植物組織、細胞、血清、血漿等樣本。其原理是脂肪酶（Lipase, LPS）催化油酯水解為脂肪酸，以銅皂法測定脂肪酸的生成速率，以計算出脂肪酶（Lipase, LPS）的活性。

樣品製備: 

建議您使用新鮮樣品。若不立即測定，樣本可在-80℃保存一個月。測定時應控制解凍的溫度和時間。室溫解凍時，樣品應在4小時內解凍。

1. 動物和組織：秤取0.1g組織，加入1mL試劑I，冰上勻漿。 4℃，12,000 g 離心 10 分鐘，取上清液進行測定，置於冰上待測。

2. 細胞：收集10⁷將細胞放入離心管中，用冷的PBS清洗細胞，離心後棄上清；加入1mL試劑I超音波破碎細胞5min（功率20%或200W，超音波3s，間隔7s，重複30次）。在 4℃ 下以 12,000 g 離心 10 分鐘。取上清液進行測定，並置於冰上待測。

3. 血漿、血清或其他液體樣本：直接測試。注意：1.高脂肪樣本離心後，如果上清液上方有固體脂質，則必須用棉花棒或類似物將其擦掉後才能進行測量。 2. 如果需要測定樣本的蛋白質濃度，建議使用Abbkine Cat #: KTD3001Protein Quantification Kit (BCA Assay) 來測定樣本的蛋白質濃度。

Background	Lipase (LPS), also known as glycerol ester hydrolase, catalyzes the hydrolysis of triglycerides to produce free fatty acids and glycerol (or di- and mono-glycerides). LPS is widely distributed in various organisms. Elevated levels of LPS in the serum are commonly observed in pancreatitis and pancreatic cancer. CheKine™ Micro Acid Phosphatase (ACP) Activity Assay Kit provides a simple, convenient, and rapid method for detecting LPS activity, applicable to samples such as animal and plant tissues, cells, serum, and plasma. It works by having LPS catalyze the hydrolysis of oil esters into fatty acids, and by measuring the rate of fatty acid production using the copper soap method, the LPS activity can be calculated.
