DNA ligase IV inhibitor
Scr7 是一種 DNA 連接酶 IV 抑制劑(DNA ligase IV inhibitor)，最初被確定為一種抗癌劑 (anti-cancer agent)。Scr7 靶向 DNA 連接酶 IV 的 DNA 結合域(DNA binding domain)，降低其對雙鏈斷裂 (double strand breaks , DSB) 的親和力並抑制其功能。 Scr7 還抑制 DNA 連接酶 III（但不抑制 DNA 連接酶 I），儘管效率較低。用強力黴素處理細胞以誘導 Cas9 表達，用不同濃度的 Scr7 處理 24 小時。 Scr7 維持的細胞能夠進入 S/G2 期，這是 HDR 所必需的 [1]。用 Scr7 治療小鼠會影響淋巴細胞發育，因為 DNA 連接酶 IV(DNA ligase IV) 在通過 C-NHEJ16 進行 V(D)J 重組過程中編碼末端的連接中起關鍵作用。由於 Scr7 的非共價結合模式，Scr7 治療後淋巴細胞發育的缺陷是短暫的和可逆的。 Scr7 通過瞬時阻斷 NHEJ（依賴 DNA 連接酶 I 的 alt-NHEJ 除外）增強 HDR 的頻率，從而在培養細胞和小鼠中通過 CRISPR-Cas9 進行精確的基因組編輯(genome editing) [2]。
