RGFP966 is a selective inhibitor of HDAC3 with IC50 value of 80 nM [1].
RGFP966 是 HDAC3 的選擇性抑制劑，IC50 值為 80 nM [1]。HDAC3（histone deacetylase 3, 組蛋白脫乙醯酶 3）是 HDACs 家族的一種異構體，在 DNA 轉錄調控(DNA transcriptional regulation)中起重要作用。已經表明，HDACs 涉及多種生物過程，如 DNA 修復(DNA repair)、複製(replication)、轉錄(transcription)和染色質結構(chromatin structure)。許多研究表明，HDACs 是大腦中表達最高的 I 類 HDAC，負向參與學習(learning)和記憶過程(memory process) [2]。RGFP966 是一種有效的 HDAC3 抑制劑(HDAC3 inhibitor)，在濃度高達 15 μM 時對其他 HDAC 沒有抑制作用。使用底物依賴性生化試驗研究RGFP966對HDACs的抑制作用，結果表明它抑制HDAC3的IC50值為80 nM，而對其他HDACs沒有有效抑制作用[1]。當用 RGFP966 對皮膚 T 細胞淋巴瘤 (T cell lymphoma , CTCL) 細胞系進行測試時，通過抑制增加細胞凋亡的 HDAC3，細胞生長(cell apoptosis)顯著降低 [3]。在雄性 C57BL/6J 小鼠模型中，在給予 RGFP966 後在亞閾值期間對它們進行物體識別 (ORM) 和位置依賴性物體識別 (OLM) 訓練，顯著增加了新的物體偏好並增強了長期 OLM 的劑量-依賴方式[1]。
