RNase R是一種大腸桿菌外切核糖核酸酶(E. coli exoribonuclease)，表現出3’至5’核酸外切酶活性(exonuclease activity)，有效消化幾乎所有linear RNA種類。該酶不消化環狀(circular)，套索狀(lariat)或雙鏈狀(double-stranded RNA)3’突出端 （少於7個）的鏈狀RNA核苷酸。因此，這種酶非常適合研究傳統剪接產生的套索狀RNA(lariat RNA produced)，以及通過反向剪接(back-splicing)產生的circRNA。RNase R通過從細胞或RNA提取物中去除線性RNA，RNase R經由RNA-sequencing有效鑑定環狀物種。
