Western Blot等檢診及分子病毒之研究!!直接在一抗上做標誌 (另有標誌螢光蛋白 螢光dye HRP Biotin等) ，克服二抗所造成的背景值過高及Host來源不同所造成Crosstalk的現象。辣根過氧化物酶 (Horseradish peroxidase, HRP) 是一種分子量為 44,173.9 道爾頓的糖蛋白(glycoprotein)，含有 6 個賴氨酸殘基(lysine residues)，可與標記分子偶聯。當與合適的底物孵育時，它會產生標記分子的有色、螢光或發光衍生物，從而可以進行檢測和定量。 HRP 常用於偶聯物中以確定目標分子的存在。由於其單體性質以及易於生成有色產物的特性，HRP 也常用於 ELISA 和免疫組織化學等技術。LinKine™ HRP 標記試劑盒旨在直接從含有遊離氨基的蛋白質、胜肽和其他配體製備 HRP 偶聯物。本試劑盒中提供的 HRP 已預先激活，可直接用於偶聯。偶聯產物可在 4℃ 穩定保存 1 個月。應用範例：偶聯的直接標記一抗在免疫測定中無需二抗，從而省去了繁瑣的孵育和洗滌步驟，節省了時間。

| Usage notes | • The purified antibody to be labeled should ideally be in 10-50 mM amine-free buffer (e.g. MES, MOPS, HEPES, PBS), pH range 6.5 to 8.5. • Common non-buffering salts (e.g. sodium chloride), chelating agents (e.g. EDTA), and sugars have no effect on Labeling efficiency. We do not recommend performing a HRP Labeling in presence of sodium azide or adding sodium azide with your HRP conjugated antibody. Glycerol up to 50% has no effect. • The amount of antibody used for Labeling should ideally correspond to molar ratios between 1:4 and 1:1 Antibody to HRP. • Antibody concentrations in the range 0.5-4 mg/ml generally give optimal results. |

| Background | Activated Peroxidase is horseradish peroxidase (HRP) enzyme, whose native carbohydrates (sugars) have been gently oxidized with periodate to produce amine-reactive aldehyde groups. These carbonyls of Activated Peroxidase spontaneously and efficiently crosslink with primary amines on antibody or other proteins. The method is more effective than other amine-reactive chemistries, such as glutaraldehyde coupling, which often causes polymerization and a greater degree of conjugate inactivation. The pre-activated enzyme eliminates the difficulties inherent in preparing and validating activated peroxidase from scratch. |

Materials Supplied and Storage Conditions
注意：緩衝液中不得添加含有氨基(amino)的成分，否則會影響偶合效果(coupling effects)。疊氮化鈉( Sodium Azide)會破壞辣根過氧化物酶 (HRP)，偶聯緩衝液中應去除Proclin。少量BSA和明膠不會影響結果，建議使用PBS作為所需緩衝液。如果樣本中含有可能干擾標記的物質，建議以PBS取代緩衝液。具體方法如下：將樣品加入超濾管中，加入200-150 µL PBS。 12,000 g，4 ℃，離心10分鐘，棄濾液。然後再次加入PBS，12,000 g，4 ℃，離心10分鐘。離心結束後，取出超濾管內芯，置於乾淨的外管中，4,000 g、4℃、2 min 離心，收集樣本。
List of components (or storage buffer) requirements of the sample to be labeled:
Q1：濃縮後樣本濃度仍低於2 mg/mL？
A1：如果濃縮後樣品濃度仍小於2mg/mL，可以調整標記系統的體積適當，但終濃度應大於1 mg/mL。步驟(1)中生物素標記溶液的體積應為標籤系統體積的 10%。步驟(2)中可以不加入去離子水，其體積為活化的生物素溶液保持不變。您也可以根據自己的實驗進行適當的調整。
Q2：針對不同分子量的待標記樣品，如何選擇適合的純化管柱？
A2：純化管柱適用於分子量為100KD至200KD的樣品。如果樣品帶有分子量高於200KD或低於100KD時，最好再配備更合適的純化尺寸柱子。
Q3：待標記樣品的分子量與生物素相似嗎？
A3：如果樣品的分子量與生物素相似，則完成本說明對應的標記步驟後，加入 30 µL 0.5 µM NH4Cl 並在 37℃ 孵育 10 分鐘淬滅遊離生物素，無需純化步驟。
Q4：不同分子量的分子有不同的偶合效果嗎？
A4：對於分子量小於5KD的分子，偶聯效果較差。您可以增加分子數量適當地。
