SDS-PAGE Protein Sample Loading Buffer (5X)

SDS-PAGE Protein Sample Loading Buffer (5×) 是一種改良的 5 倍濃縮蛋白上樣緩衝液(modified 5-fold concentrated protein loading buffer)，以溴酚藍(bromophenol blue)為染料。本上樣緩衝液使用穩定性更好、還原效果更好、無難聞氣味的還原劑(reducing agent)代替傳統的DTT或2-ME(2-mercaptoethanol)。用於常規還原SDS-PAGE蛋白樣品的上樣。

檢測步驟

Assay Procedure 1. SDS-PAGE Protein Sample Loading Buffer (5×) was dissolved at room temperature or no more than 37℃ water bath, then was stored immediately at room temperature to avoid being placed in the water bath for a long time. 2. According to the ratio of adding 1 µL SDS-PAGE Protein Sample Loading Buffer (5×) to every 4 µL protein sample, mix the protein sample and SDS-PAGE Protein Sample Loading Buffer (5×). 3. Heat at boiling water bath for 5-10 min to fully denature the protein. After cooling to room temperature, load the sample directly into the sample hole of the SDS-PAGE gel. 4. Electrophoresis is usually stopped until the blue dye reaches near the bottom end of the gel.

1. SDS-PAGE蛋白樣品上樣緩衝液（5×）在室溫或不超過37℃水浴中溶解，然後立即在室溫下保存，避免長時間放置在水浴中。

2. 依照每 4 µL 蛋白質樣本加入 1 µL SDS-PAGE Protein Sample Loading Buffer (5×) 的比例，將蛋白質樣本與 SDS-PAGE Protein Sample Loading Buffer (5×) 混合。

3. 沸水浴加熱5-10分鐘，使蛋白質充分變性。冷卻至室溫後，將樣品直接裝入 SDS-PAGE 凝膠的加樣孔中。

4. 通常停止電泳直至藍色染料到達凝膠底端附近。
