抗原修復(Antigen retrieval)
組織中部分抗原(Antigen)在甲醛或多聚甲醛固定過程中，發生了蛋白之間交聯及醛基的封閉作用，從而失去抗原性。 通過抗原修復(Antigen retrieval)，使得細胞內抗原決定族重新暴露，因此提高抗原檢測率。修復方法(Retrieval) 從強到弱一般分為三種，高壓修復、微波修復、酶修復。高壓及微波修復易造成組織損失並損害組織完整性和形態。

IHC/ICC 免疫組織化學染色步驟-冷凍，石蠟切片和細胞塗片

1.組織切片脫蠟 (Deparafinize) 和水合(hydrate )通過二甲苯 (xylene)或其他清除劑和graded alcohols使組織切片脫蠟和水合。注意事項: 對於冷凍切片(frozen sections)或細胞塗片(cell smears )使用 unfixed, acetone fixed 或適當的 fixative for the antigen 。細胞塗抹(cell smears )使用detergent – Triton or NP40 (0.1-0.2% in PBS for 10 min使滲透性。Tween20，皂甙，Digitonin或類似於leucoperm（在PBS中0.2-0.5％，持續10-30分鐘）適用於細胞質或質膜(plasma membrane)抗原。
2.用蒸餾水(distilled或去離子水(deionized water)洗滌2-3次。
3.在室溫下用過Peroxidase Block (clear cap) 切片/細胞塗片反應5-10分鐘。用蒸餾水(distilled water)洗3次。注意事項：antigen retriever （胰蛋白酶AR-6541，Pronase AR-6542，胃蛋白酶AR-6543，檸檬酸鹽緩衝液AR-6544，緩衝液(with EDTA pH 8.5 AR6545，Tris緩衝液pH 10 AR-6546），它可用於這一步。
4.用PBS或Tris saline buffer（含0.02-0.05％非離子洗滌劑，Triton X100，Tween 20或NP-40）洗滌載玻片3次。
5.切片/細胞塗片於Protein blocking solution (blue cap)，在室溫下孵育5-10分鐘。
6.Incubate sections/cell smear with primary antibody（未提供，僅提供緩衝液用於稀釋）20-30分鐘。該提供的一抗稀釋緩衝液(primary antibody dilution buffer)也可用作陰性對照。
7.用PBS洗滌載玻片5-7次。在室溫下與One-Step HRP polymer (brown cap)孵育20-30分鐘。
8.用PBS或TBS洗滌載玻片5-7次，建議使用0.025-0.05％非離子型去污劑 without
sodium azide（Triton x-100或Tween-20或NP-40）洗滌。注意事項：過氧化物酶試劑被疊氮化鈉 (sodium azide)破壞，應避免在所有緩衝液和試劑中使用。
9.用去離子水(deionized)或蒸餾水(distilled)洗2-3次。
10.在試管中，加入1 ml Reagent BS和50μl試劑C（DAB chromogen）混合 （即用型DAB試劑限7-8小時內使用完。）
11.在組織載玻片上加入幾滴即用型DAB試劑 (步驟10)，在室溫下等待6-10分鐘。 （更高溫度37°C也可用於DAB chromogen，但孵化時間應由個別實驗室確定
12.用PBS洗滌5-7次，然後用離子水(deionized)或蒸餾水(distilled)沖洗。
13.與DAB兼容的複染劑（未提供）孵育30-60秒。We prefer hematoxylin。
14.用蒸餾水(distilled )/去離子水(deionized water)清洗載玻片。
15.用適當的封片劑封片，例如SuperImmunoHistoMountTM（BV Cat.No:1219-30）
