抗原修復(Antigen retrieval)
組織中部分抗原(Antigen)在甲醛或多聚甲醛固定過程中，發生了蛋白之間交聯及醛基的封閉作用，從而失去抗原性。 通過抗原修復(Antigen retrieval)，使得細胞內抗原決定族重新暴露，因此提高抗原檢測率。修復方法(Retrieval) 從強到弱一般分為三種，高壓修復、微波修復、酶修復。高壓及微波修復易造成組織損失並損害組織完整性和形態。
產品介紹:
UNI-TRIEVE不需要煮沸或暴露於高溫下，因此不需要使用會造成組織損失並損害組織完整性和形態的壓力鍋或微波爐。使用簡單的水浴方式，在30分鐘內僅需60-75攝氏度。 完全不含背景 ( Background -Free)，不同於檸檬酸鹽緩衝液 (citrate buffer)，高pH緩衝液 (high pH buffers)，高壓鍋 (pressure cookers) 或微波爐 (microwaves)，Uni-Trieve沒有觀察到背景或組織損傷/損失 (tissue damage/ loss)。UNI-TRIEVE是一種溫和的Retrieval Solution ，可應用於所有人類和所有動物組織以及細胞製劑（細胞離心塗片，塗片）。UNI-TRIEVE也應用於需要蛋白水解酶(proteolytic enzyme)消化的組織和抗體的理想替代品。例如用蛋白酶 ( pronase)，胰蛋白酶(trypsin)和胃蛋白酶(pepsin enzymes)消化組織中的細胞角蛋白(cytokeratin)抗體等需要酶處理的抗體。只需在70oC UNI-TRIEVE中孵育30分鐘即可代替酶消化。Uni-Trieve溶液37至40oC Over-night，適用於所有膜(membrane)，胞質(cytoplasmic)和核標記(nuclear markers)，特別適用於熱敏感組織(retrieving heat-sensitive tissues)，例如腦(brain)，脂肪組織(fatty tissues)和CD標記(CD markers)。一些核抗原(nuclear antigen )，例如Estrogen Receptor (ER)，progesterone receptor (PR)，ki-67等，需要在75oC下1小時或在80°C下30分鐘。(樣品建議反應溫度及時間，請參考原廠操作手冊)
