ToxOut™ Rapid Endotoxin Removal Kit
polymyxin B去除內毒素之原理:
Polymyxin B（PMB）是從多粘芽孢桿菌中分離出來的一種強陽離子環抗生素，是目前發現的唯一一種能特異性親和吸附內毒素(脂多糖)的抗生素。將Polymyxin B通過共價偶聯的方式固定在瓊脂糖珠上，利用Polymyxin B能夠特異性親和內毒素的特性，除去溶液和生物製品中的內毒素。Biovision提供一系列內毒素試劑商品；其中ToxOutTM快速內毒素去除試劑盒 (貨號K2501) 中，有效消除溶液中的內毒素<0.05EU / ml，高結合容量(Binding Capacity)：高達up to 9 x 108 EU/ml resin。高效樣品回收率(Sample Recovery) 蛋白質溶液樣品回收〜90％。親和性基質polymyxin B，50％漿液溶於20％乙醇中。Columns Content: 5 x 100 µl ToxOutTM Rapid Endotoxin Removal Agarose (Cat. 快速內毒素去除 (即用型離心柱)
包含 100uL 快速內毒素去除瓊脂糖。任何時候都不要讓樹脂變乾。使用後儲存在含有 20% 乙醇的內毒素平衡緩衝液中。如果購買單獨的緩衝液（內毒素去除平衡緩衝液 (ab278802)、快速內毒素去除再生緩衝液 (ab269281)、快速去除洗滌緩衝液 (ab278801)），每根管柱最多可重複使用 5 次。
內毒素介紹:
內毒素(Endotoxins )是位於革蘭氏陰性細菌(gram negative bacteria)外膜(outer membrane )中的脂多醣（LPS）複合物。單個大腸桿菌含有大約2百萬LPS之分子（2-20fg /細胞）。在實驗過程中，大量的內毒素被脫落並且可以容易地污染實驗室器皿，緩衝液及下游產品。In vitro，內毒素在基於細胞的研究中引起各種問題；In vivo，內毒素可引起各種副作用，包括宿主生物體中的炎症反應(inflammatory response)，器官衰竭(organ failure)或膿毒性休克(septic shock)。內毒素對細胞的培養具有有害的影響，並且可導致不可再現的或陰性實驗的產生數據。
簡單操作程序:
The kit quickly and effectively eliminates endotoxins to < 0.05 EU/ml and is ideal for processing small-scale solution sample (0.1-0.5 mL)
將管柱進行以下反應 (加入-離心)
—-移去管柱中下面的栓，並放至ToxOut™Endotoxin-free Collection Tube. Spin離掉 storage solution.
—-加入Equilibration(total two times)溶液並離心-> 加入Regenerate溶液並離心-> Wash(total 3 times)-> 加入Equilibration (total two times) 溶液並離心->
—- Place column in a new ToxOutTM Endotoxin-free Collection tube
—- 加入sample (0.1 – 0.5 ml)重力流動(3-5 times), 將cap 及 plug裝回管柱
—- room temperature or 4 ̊C for 1 hour
—-離心收集去除完內毒素之Sample
Rapid Endotoxin Removal Spin Column Ready to use as supplied.
Contains 100uL of Rapid Endotoxin Removal Agarose. Do not let the resin dry out at any time. Store in Endotoxin Equilibration Buffer containing 20% ethanol after use. Each column can be re-used up to 5 times, providing the separate buffers are purchased (Endotoxin Removal Equilibration Buffer (ab278802), Rapid Endotoxin Removal Regeneration Buffer (ab269281), Rapid Removal Wash Buffer (ab278801)).
重柱若重覆使用Pace plug and cap back and store the column in 200 µl ToxOutTM Endotoxin Removal Equilibration buffer with 20% ethanol at 4 ̊C
