總抗氧化能力(The total antioxidant)由被測物中各種抗氧化物質(antioxidant substances )和抗氧化酶(antioxidant enzymes )組成。 ABTS法 (ABTS method) 是應用最廣泛的間接檢測方法(indirect detection method),適用於測定親水性(hydrophilic)和親脂性物質(lipophilic substances)的抗氧化能力。 CheKine™ Micro ABTS自由基清除能力檢測試劑盒 (CheKine™ Micro ABTS Free Radical Scavenging Capacity Assay Kit) 提供了簡單、快速的ABTS自由基清除活性測定方法,適用於植物組織 (plant tissues)、紅酒 (red wine)等液體樣本。其原理是ABTS被氧化生成穩定的藍綠色陽離子自由基ABTS+,ABTS+可溶於水或酸性乙醇介質,並在734 nm處具有最大吸收。將待測物質加入ABTS+溶液後,樣本中的抗氧化成分與ABTS+反應,導致反應體系褪色。在734 nm處測量吸光度的變化,並透過與維生素C參考體系進行比較來量化待測物質的抗氧化能力。

樣品製備:

1. 植物樣品製備:秤取約0.1 g樣品,加入1 mL萃取緩衝液,冰上勻漿。 4℃下以10,000 g離心10 min,收集上清液,置於冰上保存以備後續分析。
2. 液體樣品(如紅酒、果汁等):吸取100 μL樣品溶液加入900 μL萃取緩衝液中,渦旋混勻。 4℃下以10,000 g離心10 min,收集上清液,置於冰上保存以備後續分析。
檢測步驟
1. 將微孔板讀數儀或可見分光光度計預熱30分鐘以上,將波長調至734nm,用去離子水將可見分光光度計歸零。 2. 操作步驟(以下操作皆在96孔板或微型玻璃比色皿中進行):
