DPPH自由基 (DPPH radical) 是一種非常穩定的氮中心自由基 (nitrogen-centered free radical),是樣品抗氧化能力(antioxidant capacity)的重要指標之一,廣泛應用於抗氧化食品(antioxidant foods)、保健品 (health supplements) 和藥品 (pharmaceuticals) 的研究。 CheKine™ 微量DPPH自由基清除能力檢測試劑盒( CheKine™ Micro DPPH Free Radical Scavenging Capacity Assay Kit, 貨號KTB1092) 提供了一種簡單、快速的DPPH自由基清除活性 (DPPH radical scavenging activity) 測定方法,適用於植物組織(plant tissues)、紅酒(red wine)等液體樣本。其原理是DPPH自由基具有未配對電子(unpaired electron),使其乙醇溶液(ethanol solution)呈現紫色,並在515 nm處有強烈吸收。當存在抗氧化劑時,DPPH自由基被清除,導致溶液顏色褪去,515 nm處的吸光度降低。在一定範圍內,吸光度的變化與自由基清除(radical scavenging)程度成正比。
樣品製備:
1. 將微孔板讀數儀或可見分光光度計預熱30分鐘以上,將波長調至515nm,以無水乙醇將可見分光光度計歸零。 2. 操作步驟(以下操作皆在96孔板或微型玻璃比色皿中進行):

注意:每個測試孔應設置相應的對照孔;陽性對照孔和空白孔只需測量1-2次。不同樣本清除DPPH自由基的能力可能有顯著差異。建議在進行正式實驗前,先用2-3個預期差異較大的樣品進行預實驗。若清除率低於5%,可適當增加樣品量,例如增加乾燥樣品的重量或液體樣品的體積。若清除率超過90%,可用萃取緩衝液進一步稀釋樣品,並將計算結果乘以稀釋倍數,或減少萃取所使用的樣品量。


注意事項
1. 盡量避免使用在酸性條件下會變紅或近紅色的試劑,因為它們可能會干擾本試劑盒的檢測結果。
2. 請勿添加Tween、Triton和NP-40等洗滌劑,或DTT(二硫蘇糖醇)和巰基乙醇等還原劑,這些物質會影響樣本的氧化還原反應。
3. 如果液體樣本呈鹼性,則應在測試前使用萃取緩衝液將其稀釋至酸性。
4. 不同樣本清除DPPH自由基的能力可能有顯著差異。如果要比較不同樣本的DPPH自由基清除能力,建議使用同一批次的等量樣本進行測試。
5. 建議樣品在同一天進行萃取和測試。
