二級抗體 (Secondary antibodies) 應用:
二抗(Secondary antibodies)是針對免疫球蛋白(immunoglobulin , Ig)分子的多克隆(polyclonal) 和單克隆抗體(monoclonal antibodies)。它們通常與抗原特異性一抗(specific primary antibody) 一起使用,並且通常被標定酵素或螢光物質以幫助檢測。二抗可用於多種應用,例如ELISA、免疫印跡(Immunoblotting)、流式細胞儀(Flow Cytometry)、免疫組織染色(Immunohistochemistry)、免疫細胞化學(Immuocytochemistry)。二級抗體通常用酶或螢光染料標記,這使得能夠通過比色,化學發光和螢光技術進行檢測。例如,酶辣根過氧化物酶(enzymes horseradish peroxidase , HRP)和鹼性磷酸酶(alkaline phosphatase , AP)通過比色和化學發光平台在ELISA和蛋白質印跡應用中是有價值的。螢光蛋白諸如螢光素(fluorescein , FITC),藻紅蛋白(phycoerythrin , PE)和花青(cyanine , CY)的螢光染料在螢光的應用中是必不可少的,包括流式細胞術,免疫螢光顯微術和多重測定。二抗允許標記和檢測的靈活性以及通過信號放大,可增加的靈敏度。
