產品介紹: 困難梭菌谷氨酸脫氫酶（GDH）多株抗體-生物素標記, 其免疫原自困難梭菌谷氨酸脫氫酶進行雞隻免疫。在弗氏佐劑中進行多次免疫後，收集蛋，並將IgY純化。使用固定在固體支持物上的免疫原將困難梭菌谷氨酸脫氫酶進行免疫親和純化。

使用雞抗體的好處：

雞非哺乳類動物，和哺乳類抗原有高親和性，且避免和哺乳類抗原產生cross-reaction。
為最人道的多株抗體產製方法。只需從雞蛋萃取蛋白，取樣方便，不傷害動物。
每隻雞可以產生的抗體量約3 g IgY／月，是兔子的10-20倍多。且產製速度比起兔子快上許多，最快25天即可取得高力價（titer）抗體。
雞IgY的Fc region和哺乳類IgG的不同：
- 可降低對哺乳動物的背景值。
- 不會活化哺乳類補補體系統。
- 不會與哺乳類Fc recentors結合。
- 無法和常規的protein A（Staphylococcus aureus）、protein G（Streptococcus sp.）、或protein L（Peptostreptococcus magnus）等免疫球蛋白－蛋白結合反應。
由於IgY取得方便，雞蛋可以儲存較長的時間，並可純化取得較佳的titer。
相對於兔子，雞的開銷較便宜。
IgY有以下數種和哺乳類IgG類似的特徵：
- 為二價體。
- 可由papain分解成divalent Fab fragment。
- 適用酵素、生物素（biotin）、金粒子等標定物。

Procedure	Chickens were immunized with highly purified Glutamate Dehydrogenase (GDH) purified from Clostridium difficile. After multiple immunizations in Freund’s adjuvant, eggs were collected and the IgY purified. Anti-Clostridium difficile GDH was immunoaffinity purified using the immunogen immobilized on a solid support and conjugated to Biotin.
