產品介紹: LB培養基（Miller）（Lysogeny broth，LB），是微生物學實驗中最常用的營養性培養基，用於培養大腸桿菌等細菌之生長和維持。分為液態培养基和加入瓊脂製成的固態培養基。Minimum QC organisms: Escherichia coli DH5 a(ATCC® 53868)。LB Broth(Miller)培養基用於分子微生物學程序中使用的大腸桿菌菌株的生長和維持，營養豐富，旨在用於重組菌株純培養物的生長。大腸桿菌在LB培養基中生長至對數後期(late log phase)。一些vectors載體無需選擇性擴增(selectively amplified) 即可複製至高拷貝數(high copy numbers)。一些載體不能自由複製，需要選擇性擴增。可以加入氯黴素(Chloramphenicol)以抑制宿主合成並防止細菌染色體複製。LB Broth(Miller)培養基含有高含量的氯化鈉(sodium chloride)，有助於維持plasmids。如果使用lambda噬菌體 (temperate bacteriophages)，建議添加2g / l的硫酸鎂(magnesium sulphate)以促進噬菌體吸收(phage absorption)。如果需要，可以在滅菌後對培養基進行無菌葡萄糖(glucose)補充。

培養基使用方法: 將25克培養基溶於1升純淨水中。如有必要，經常攪拌加熱以完全溶解Medium。在121℃下高壓滅菌15分鐘。等冷去後，最終pH：7.3 ± 0.2, 25℃分裝於Petri dish培養皿中。

檢驗方法：檢體經系列稀釋後，以平板計數培養基培養及計數之方法。 2.1. 工作環境：工作平台須寬敞、潔淨、光線良好，操作平台光度為 100 呎燭光以上，密閉室內換氣良好，儘可能沒有灰塵及流 動空氣。每 15 分鐘落菌數不得超過 15 CFU/培養皿。更多資料來自 FDA生菌數之檢驗 (參考方法)
