用血清(sera)或蛋白質封閉劑(protein blocking reagent)，進行免疫染色前之封閉(Blocking)，可防止抗體與組織或Fc受體之非特異性結合(non-specific binding)。從理論上講，任何不與靶抗原結合的蛋白質都可以用於封閉(Blocking)。血清(sera)是一種常見的阻斷劑，因為它含有與非特異性位點結合的抗體。建議使用與二抗物種匹配的血清。當使用不同物種的第二抗體進行多次染色時，可能有必要使用兩種第二抗體物種的封閉血清(blocking sera)。蛋白質如BSA或酪蛋白(casein)之類的也可以用於阻斷非特異性抗體(non-specific antibody binding)的結合，並且通過這些蛋白質，無需使試劑與第二抗體的種類相匹配。專門的封閉緩衝液也經常用於封閉非特異性抗體結合。

Normal serums

正常血清(Normal Mouse serums)，來自非免疫動物宿主的正常血清。正常血清可應用於免疫反應中之blocking，建議PBS稀釋至5％-20％（v / v）正常血清作為封閉劑(blocking reagent)，以減少非特異性結合(non-specific binding)的背景或用於大多數immunoassay applications的對照。亦可在加入一級抗體之前將Blocking reagent先進行反應。最佳工作稀釋度應由研究者通過實驗確定。建議的起始稀釋度如下：IHC-p（1：10-20），IF / ICC（1：10-20）和ELISA（1：100）。

Optimal working dilutions should be determined experimentally by the investigator. Suggested starting dilutions are as follows: IHC-p (1:10-20), IF/ICC (1:10-20) and ELISA (1:100).

• Preparation method

Normal mouse serum is processed from blood collected from non-immunized normal adult mouses. Hemolysis is minimized by employing proprietary techniques during the blood collection and serum extraction processes.
