許多質體DNA含有編碼六個組氨酸（His）殘基組成的N-末端標籤的DNA，及擴展的extended multiple cloning sites多克隆位點。 表達的重組蛋白上的6xHis標籤允許有效偶聯(coupling to) 至Ni ++親和樹脂(Ni++ affinity resins)並通過單步色譜法(single step chromatography)純化。與其他蛋白質標籤(protein tags)的情況一樣，該多組氨酸標籤(polyhistidine tag)可以在酶（例如凝血酶(thrombin)和腸激酶(enterokinases)）識別的位點處被切割以分離目的蛋白質。
