關於膠原（Collagen）：
膠原蛋白是動物結締組織重要的蛋白質,在哺乳動物中的含量尤為豐富，佔有機體蛋白質百分之25%-35%含量。膠原是細胞外最重要的水不溶性纖維蛋白, 是構成細胞外基質（extracellular matrix, ECM）的骨架。膠原在細胞外基質中形成半晶體的纖維,給細胞提供抗張力和彈性,並在細胞的遷移和發育中起作用。膠原蛋白的基本結構單位是原膠原(tropocollagen), 原膠原肽鏈的一級結構具有(Gly-x-y)n重複序列, 其中x常為脯氨酸(Pro), y常為羥脯氨酸(Hypro)或羥賴氨酸(Hylys)。Hylys殘基可發生糖基化修飾,其糖單位有的是一個半乳糖殘基(Gal), 但通常是二糖(Glu-Gal-),膠原上的糖所占的量約為膠原的10%。原膠原是由三條α-肽鏈組成的纖維狀蛋白質, 相互擰成三股螺旋狀構型, 長300nm, 直徑1.5nm。目前已發現29種不同組織中不同類型的膠原; 不同類型的膠原定位於體內的特定組織,也有2-3種不同的膠原存在於同一組織中。而機體內90%的膠原為I、II、III、IV型。
膠原蛋白(Collagen)是細胞外空間的主要結構蛋白，也是 ECM 中最豐富的蛋白質。膠原蛋白分為兩類 – 原纖維（ fibril) I、II、III、V 型）和非原纖維(non-fibril)（IV、VI 型）。 I 型膠原蛋白(Type I collagen)是最豐富的膠原蛋白，幾乎在所有結締組織中表達，包括皮膚(skin)、肌腱(tendon)和骨組織(bone tissue)。它也是間隙基質(interstitial matrix)的主要成分。 I 型膠原蛋白突變與一系列疾病有關，包括成骨不全症(osteogenesis imperfecta)和埃勒斯-當洛斯症候群(Ehlers–Danlos syndrome)。 I 型膠原蛋白由兩條 α1(I) 鏈和一條 α2(I) 鏈組成。
| Description | Collagen is the main structural protein in the extracellular space and is the most abundant protein in the ECM. Collagens are divided into two classes – fibril (types I, II, III, V) and non-fibril (types IV, VI). Type I collagen is the most abundant collagen and is expressed in almost all connective tissues including skin, tendon, and bone tissue. It is also the predominant component of the interstitial matrix. Type I collagen mutations are associated in a range of diseases including osteogenesis imperfecta and Ehlers–Danlos syndrome. Type I collagen consists of two α1(I) chains and one α2(I) chain. |
