產品介紹

EZ Cap™ Cy5 螢火蟲螢光素酶 mRNA (5-moUTP) 一旦進入最初從螢火蟲 Photinus pyralis 中提取的細胞就會表達螢光素酶蛋白。該酶催化 ATP 依賴性 D-螢光素氧化並導致在約 560 nm 波長處產生化學發光。螢火蟲螢光素酶是一種常用的生物發光報告基因,用於基因調控和功能研究。適用於 mRNA 傳遞( mRNA delivery)、翻譯效率(cell viability)、細胞活力(cell viability)和體內成像等檢測。EZ Cap™ Cy5 螢火蟲螢光素酶 mRNA (5-moUTP) 的濃度為 1 毫克/毫升。它由 EZ Cap™ Reagent AG (3′ OMe)(貨號 B8178)共轉錄封端,可高效生成帽 1 結構。 Cap 1 結構更適合哺乳動物系統,轉錄效率高於 Cap 0 結構(ARCA 和 mCap)。 EZ Cap™ Reagent AG (3′ OMe) 獲得了僅以正確方向插入的能力,導致形成的 mRNA 的翻譯效率是使用 EZ Cap™ Reagent AG 啟動的 mRNA 的兩倍。添加 5-moUTP (5-Methoxy-UTP) 和 poly(A) 尾巴可抑制 RNA 介導的先天免疫激活並增加 mRNA 在體外和體內的穩定性和壽命。 Poly(A) 尾在提高翻譯起始效率方面也起著重要作用。 Cy5 是一種合成的紅色螢光染料,最大激發和發射波長分別為 650 nm 和 670 nm。 Cy5-UTP 和 5-moUTP 在轉錄時以 1:3 的比例使用。按此比例進行替換會產生易於可視化且仍可在細胞培養物中翻譯的 mRNA。粗略地說,翻譯效率和 Cy5-UTP 替換之間存在反相關。所有的修改都是為了模仿一個完全加工的成熟 mRNA。 EZ Cap™ Cy5 螢火蟲螢光素酶 mRNA (5-moUTP) 是觀察 mRNA 傳遞(mRNA delivery)、定位(localization)、翻譯( translation a)和其他行為的理想產品。

General tips Dissolve it on ice and take care to prevent RNase contamination degradation. Avoid repeated freezing and thawing as much as possible. Do not vortex. For the first time, it is gently centrifuged and divided into several parts for stand-alone use. Use RNase-free reagents and consumables, using appropriate RNase-free technology. It can not be added to the serum-containing medium until it is mixed with the transfection reagent.
