Human peripheral blood monocyte cells (PBMC) were isolated from whole blood and cultured in serum-free RPMI 1640 for 1 hour at 37˚C. Non-adherent cells were removed, and the remaining monocytes were cultured in RPMI 1640 supplemented with 10% FBS, 2 ng/ml GM-CSF (Cat. No. Z100275), and 10 ng/ml IL4 (Cat. No. Z100545). Mature dendritic cells were induced by addition of 20 ng/ml TNFa (Cat. No. Z100855) on day 6 and cultured for 24 hours. The arrow shows matured dendritic cell morphology as well as interaction with and activation of T cells.
