To detect botulinum toxin E in unknown samples using an indirect capture ELISA
肉毒桿菌毒素（BTX）是由肉毒桿菌和相關物種產生的神經毒性蛋白質。有八種類型的肉毒桿菌毒素，名為A-H型。Tetracore提供肉毒桿菌毒素A（Botulinum Toxin A）、肉毒桿菌毒素A&B（Botulinum Toxin A&B）、肉毒桿菌毒素C（Botulinum Toxin C）、肉毒桿菌毒素E（Botulinum Toxin E）、肉毒桿菌毒素F（Botulinum Toxin F）等ELISA檢測試劑盒，可偵測環境中的肉毒桿菌。產品具有(+/-) 的ELISA會有positive和negative的capture antibody行行交錯，negative可直接對照上行的positive結果，在混合物的sample中可得到較佳的結果。【注意事項】本類套組不含標準品（需自備），Not for diagnostic use of clinical samples。
產品描述：
Botulinum Toxin E, Pre-Coated (+/-) 用於檢測環境樣品中的特定試劑（和模擬物）。本產品不適用於臨床樣本的診斷用途。加/減 (+/-) ELISA 格式在正常、相同物種抗體的負排上方使用一排陽性捕獲抗體。可以從頂部結果中減去較低的結果以產生調整後的結果。這是複雜樣品基質的首選方法。注意事項: pre-coated（+/-）肉毒桿菌毒素B BioThreat Alert ELISA試劑盒（TC-4037-002）無須再進行陽性捕獲抗體(Positive Capture Antibody)及陰性捕獲抗體 (Negative Capture Antibody)之coated動作，試劑中亦不包括陽性捕獲抗體(Positive Capture Antibody)及陰性捕獲抗體 (Negative Capture Antibody)。此產品不包括肉毒桿菌毒素B之標準品。需經由儀器測量讀值 ELISA儀器設備

Buffer配製
PBS Coating Buffer：1升去離子水或過濾水中加入一包PBS (0.01 M phosphate buffered) 。使用電磁加熱攪拌器的磁力攪拌棒混合溶液直至溶液溶解。以0.22μm過濾。在4°C下儲存長達一個月。PBST (Phosphate Buffered Saline with Tween-20)：向800mL PBS包衣緩衝液中加入0.8mL Tween-20，使溶液混合直至溶解。在4°C下儲存長達一個月。ELISA稀釋/封閉緩衝液 (ELISA Dilution/Blocking Buffer)：對於每個使用的平板，將5克乾脫脂乳加入90mL PBST中。使用攪拌棒和攪拌板使混合物進入溶液。使用氫氧化鈉或等效物將pH調節至7.4。添加PBST使總體積達到100 mL。在4°C下儲存最多3天，或在-20°C下等分儲存最多一個月。注意：對於長期儲存，可以在緩衝液中加入0.001％ thimerosal（Sigma T5125）。，製備1％thimerosal在去離子水或過濾水中的溶液，然後每1升緩衝液加入1mL該混合物，產生0.001％溶液。
