產品介紹: N-煙酰胺甲基轉移酶（NNMT）（E.C.2.1.1.1.1）使用S-腺苷甲硫氨酸（SAM）作為供體催化煙酰胺，吡啶和其他類似物的N-甲基化，產生1-甲基煙酰胺（MNA）。NNMT在代謝途徑的調控中起重要作用，並在幾種癌症，神經退行性疾病，肥胖症和糖尿病中顯著高表達，表明它是治療的潛在分子靶點。 Biovision的NNMT抑制劑篩選試劑盒利用SAM作為甲基供體和煙酰胺作為底物。產生S-腺苷高半胱氨酸（SAH）和1-甲基煙酰胺的NNMT甲基化煙酰胺。 SAH通過SAH水解酶水解形成高半胱氨酸，其游離巰基使用Thiol檢測探針檢測，產生增強的螢光信號，可以在Ex / Em = 392 / 482nm測量。在NNMT抑制劑存在下，酶活性被抑制，導致螢光降低。該測定試劑盒是一種簡單，靈敏和快速的篩選NNMT潛在抑制劑工具。
