【EdU DNA合成】直接檢測DNA合成或細胞週期S期合成的方法

測量DNA增殖的最準確方法是直接測量DNA合成,最常用的方法是使用基於抗體的核苷類似物(nucleoside analog)溴脫氧尿苷(bromo-deoxyuridine , BrdU)檢測。EdU(5-乙炔基-2′-脫氧尿苷)是胸腺嘧啶核苷的替代品,它是BrdU的替代品,它也用於DNA增殖檢測,比BrdU檢測更簡單,更快。在EdU染色中,EdU被樣品中的細胞摻入新合成的DNA中。然後加入螢光疊氮化物,例如Green Fluorescence。螢光疊氮化物小到足以在天然組織和DNA中自由擴散,並且在“點擊”化學反應中共價交聯到EdU。與使用BrdU相比,EdU染色的主要優點是: EdU染色無需進行苛刻的DNA水解/ DNA變性步驟(與BrdU分析不同,在BrdU分析中,它用於使BrdU抗體進入DNA中的BrdU。EdU染色比BrdU染色更快,步驟更少。
