膠原纖維(Collagen fibers)是結締組織(connective tissue)中分佈最廣、最豐富的纖維，廣泛存在於各種器官中，特別是在皮膚(skin)、鞏膜(sclera)和肌腱(tendons)中。 I型膠原纖維(Type I collagen fibers)主要存在於骨骼(bone)、皮膚(skin)和肌腱(tendons)中；II型膠原纖維(Type II collagen fibers)主要存在於軟骨(cartilage)中；III型膠原纖維(Type III collagen fibers )主要存在於胚胎組織( embryonic tissues )、成人血管(adult blood vessels )和胃腸道(gastrointestinal tract)中；IV型膠原纖維(Type IV collagen fibers )主要存在於基底膜( basement membrane)中。膠原纖維的染色方法包括范吉森染色(Van Gieson staining)、馬松三色染色(Masson trichrome staining)、維多利亞藍膠原纖維染色(Victoria blue collagen fiber staining)和天狼星紅染色(Sirius red staining)。在偏振光顯微鏡下，膠原纖維呈現正單軸雙折射。與天狼星紅染色結合使用時，雙折射現象會增強，從而提高分辨率，並有助於區分不同類型的膠原纖維。
