Firefly Luciferase mRNA with Cap 1 structure, providing higher transcription efficiency and enhanced stability.

EZ Cap™ 螢火蟲螢光素酶 mRNA (EZ Cap™ Firefly Luciferase mRNA) 進入細胞後會表現螢光素酶蛋白,該蛋白最初是從螢火蟲 Photinuspyralis 中提取的。此酵素催化 ATP 依賴性 D-螢光素氧化並導致在約 560 nm 波長處產生化學發光。螢火蟲螢光素酶 (Firefly Luciferase)是一種常用於基因調控和功能研究的生物發光報告基因。它適用於 mRNA 遞送 (mRNA delivery)、翻譯效率(translation efficiency)、細胞活力(cell viability)和體內成像( in vivo imaging)等檢測。目前對mRNA加帽的方法有兩種:一是共轉錄法(co-transcription method),即在轉錄過程中添加Cap類似物。另一種是酵素法封蓋(enzymatic Capping)。轉錄後,Cap0 由痘苗病毒加帽酶 (VCE)、GTP 和 S-腺苷甲硫氨酸 (SAM) 進行加帽。然後透過 2´-O-甲基轉移酶和 SAM 將 Cap0 生成為 Cap1。 Cap1 封端也可以透過在一步過程中加入 VCE、2´-O-甲基轉移酶、GTP 和 SAM 來進行。 Cap 1結構比Cap 0結構更適合哺乳動物系統,並且具有更高的轉錄效率。添加 Poly (A) 尾巴可提高 mRNA 在體外和體內的穩定性和壽命。 Poly (A) 尾在提高翻譯起始效率方面也扮演重要角色。
