DYKDDDDK 標籤(DYKDDDDK-tag s)系統利用短的親水性 8 胺基酸胜肽( 8- amino-acid peptide)，該胜肽與目標蛋白融合1。 DYKDDDDK 勝肽與抗體 M1 結合。結合是鈣依賴方式(calcium-dependent manner)還是獨立方式(independent)仍有爭議。此系統的缺點是單株抗體純化基質不如其他基質穩定。一般來說，小標籤可以用特異性單株抗體檢測。為了改進 DYKDDDDK 標籤的偵測，開發了 3x (DYKDDDDK) 系統。這種三串聯 DYKDDDDK 抗原決定位是親水性的，長度為 22 個胺基酸，可以檢測高達 10 fmol 的表達融合蛋白。DYKDDDDK 標籤可以透過腸激酶(enterokinase)處理去除，腸激酶(enterokinase)對胜肽序列的 5 個 C 端胺基酸具有特異性。The solubility of this peptide in sterile water is >10 mM. Stock solution should be splited and stored at -80°C for several months.
