S3I-201 是 Stat3 的選擇性抑制劑，IC50 值為 86 μM [1]。
在體外 Stat3 DNA 結合測定(in vitro Stat3 DNA-binding assay)中，S3I-201 顯示出對 Stat3 DNA 結合活性的有效抑制，平均 IC50 為 86 μM。在 EMSA 測定中，與 Stat1 和 Stat5 相比，S3I-201 選擇性抑制 Stat3 DNA 結合活性。它抑制 Stat1-Stat3 和 Stat1-Stat1 的複合物形成，IC50 值分別為 160 和 > 300 μM。除此之外，未磷酸化、無活性的 Stat3 單體被發現可以恢復 S3I-201 抑制的 Stat3 DNA 結合活性，表明這種抑制與激活狀態無關。在 NIH 3T3/v-Src 成纖維細胞中，S3I-201 抑制 Stat3 的組成型激活並降低 pTyr-705 Stat3 水平。此外，S3I-201被發現在濃度為30-100 μM時顯著誘導具有持續活性Stat3的細胞凋亡。 S3I-201 還降低了這些細胞中細胞週期蛋白 D1、Bcl-xL 和存活的表達 [1]。
