CD45 Recombinant Protein

蛋白磷酸酶 (The protein phosphatase , PTP) 受體 CD45 是一種 I 型跨膜蛋白( type I transmembrane protein )，由一對細胞內酪氨酸磷酸酶結構域(intracellular tyrosine phosphatase domains )和可變剪接(alternative splicing)產生的細胞外結構域組成。 CD45 的催化活性是第一個磷酸酶結構域 ( first  phosphatase domain, D1) 的函數，而第二個磷酸酶結構域 ( second phosphatase domain, D2) 可以與第一個結構域相互作用並使其穩定，或募集/結合底物。 CD45 直接與抗原受體複合蛋白(antigen receptor complex proteins)相互作用或激活參與調節 T 細胞和 B 細胞抗原受體信號傳導的 Src 家族激酶(Src family kinases)。具體而言，CD45 使 Src 家族激酶 Lck ( Src-family kinases Lck )和 Fyn 在其保守的負調節( conserved negative regulatory)羧基末端酪氨酸殘基( carboxy-terminal tyrosine residues)處去磷酸化並上調激酶活性。相反，研究表明 CD45 還可以通過使正調節自磷酸化位點去磷酸化來抑制 Lck 和 Fyn。 CD45 似乎既是正調節因子又是負調節因子(negative regulator)，根據特定刺激和細胞類型傳導信號。人類白細胞(Human leukocytes )包括淋巴細胞( lymphocytes)、嗜酸性粒細胞(eosinophils)、單核細胞(monocytes)、嗜鹼性粒細胞(basophils)和中性粒細胞(neutrophils )表達 CD45，而紅細胞( erythrocytes )和血小板(platelets)不表達 CD45。
