A. 內毒素去除原理

PurKine™內毒素去除樹脂(PurKine™ Endotoxin Removal Resin) 可有效降低蛋白質樣品中的內毒素(Endotoxins)水平。內毒素(Endotoxins)，也稱為脂多醣(lipopolysaccharides)或LPS，是革蘭氏陰性細菌（Gram-negative bacteria）, 例如大腸桿菌(E. coli）的細胞膜成分(cell membrane components)。內毒素(Endotoxins)強烈影響DNA轉染至原代細胞(primary cells)和敏感培養細胞中，內毒素(Endotoxins)水平升高導致轉染效率(transfection efficiencies)急劇降低。樹脂(Resin)由90μm交聯的4％瓊脂糖珠(cross-linked 4% agarose)組成，且由修飾的多粘菌素B(modified polymyxin B)與之偶聯。該產品組合具優化(optimization)，以實現最大的蛋白質產量(protein yield)，穩定性(stability)和溶解性(solubility)。測試確認，至少重複使用五次後。性能不會降低。

PurKine™ Endotoxin Removal Resin has been designed to reduce endotoxin levels in protein samples by ≥99% while maximizing protein recovery. The Resin consists of 90μm beads of cross-linked 4% agarose, to which modified polymyxin B (PMB) has been coupled. Tests confirm that performance equals or exceeds popular polymyxin B (PMB) resins from other suppliers, and no decrease in performance occurs after at least five repeated uses.
