骨化二醇(Calcifediol )是一種前激素(prehormone)，由肝臟中維生素 D3（cholecalciferol, 膽骨化醇）的羥基化(hydroxylation)產生。然後骨化二醇(Calcidiol )在腎臟中（通過 25(OH)D-1α-羥化酶）轉化為骨化三醇(calcitriol) (1,25-(OH)2D3)，這是一種類固醇激素( secosteroid hormone )，是維生素 D 的活性形式。它也可以在腎臟中通過 24-羥基化(24-hydroxylation)轉化為 24-羥基骨化二醇(24-hydroxycalcidiol)。在醫學上，骨化二醇(calcidiol) 的血液濃度被認為是維生素 D 狀態(vitamin D status)的最佳指標。儘管專家呼籲提高不同實驗室的標準化和可重複性，但它是最敏感的測量方法。正常範圍因年齡和地理位置等多種因素而有很大差異。建議採用 20-150 nmol/L 的廣泛參考範圍，而一些研究已將低於 80 nmol/L 的水平定義為維生素 D 缺乏的指示。

Test principle:	This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with 25 Hydroxy Vitamin D(25-OH-D) protein. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to 25 Hydroxy Vitamin D(25-OH-D). Next,Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of 25 Hydroxy Vitamin D(25-OH-D) in the samples is then determined by comparing the OD of the samples to the standard curve.
