神經生長因子(NGF)是神經營養因子,神經肽主要參與某些目標神經元的生長(growth),維持(maintenance),增殖(proliferation)和存活的調節。 據報導,當嗜鉻細胞瘤細胞系PC12(pheochromocytoma cell line )暴露於神經生長因子(NGF)時,細胞通過停止細胞分裂和延長神經突(Greene和Tischler,1976)。 將細胞在含有5%胎牛血清和10%馬血清的Ham’s F12K上生長在聚賴氨酸或膠原包被的平板上。 當細胞達到對數期生長時,將新鮮培養基與10ng / mL NGF一起加入,然後每天用倒置顯微鏡觀察細胞。用NGF(10ng / mL)培養6天後PC12細胞的細胞分裂停止和分化如圖1所示。不含NGF的對照組不顯示神經突生長,細胞迅速繁殖。

除了調節神經元(neurons)生命週期(life cycle )的途徑外,NGF 還參與其中。 此外,Alpha-2-巨球蛋白 (Alpha-2-Macroglobulin, a2M) 已被確定為 NGF 的相互作用物,因此進行了結合 ELISA 測定以檢測重組人 NGF 和重組人 a2M 的相互作用。 簡而言之,將 NGF 在含有 0.01% BSA (pH 7.4) 的 PBS 中連續稀釋。 然後將 100μL 的重複樣品轉移到 2M 包被的微量滴定孔中,並在 37℃下孵育 2 小時。 用 PBST 洗滌孔並用抗 NGF pAb 孵育 1 小時,然後吸出並洗滌 3 次。 與 HRP 標記的二抗孵育後,吸出孔並洗滌 3 次。 加入底物溶液後,將孔在 37℃下孵育 15-25 分鐘。 最後,向孔中加入 50µL 終止液,並立即在 450nm 處讀數。 NGF與a2M的結合活性如圖2所示,這種作用呈劑量依賴性。

Reconstitute in 20mM Tris, 150mM NaCl (pH8.0) to a concentration of 0.1-1.0 mg/mL. Do not vortex.
STORAGE
Avoid repeated freeze/thaw cycles. Store at 2-8°C for one month. Aliquot and store at -80°C for 12 months.
STABILITY
The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37°C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.
