端粒（Telomere）是位於真核細胞染色體(eukaryotic cell chromosome)末端的一段DNA序列(DNA sequence )，由多個重複核苷酸元件（multiplerepeatingnucleotide elements, TTAGGG）串聯組成。為非轉錄的DNA( non-transcribed DNA)提供緩衝，還可以保護染色體( chromosome)末端不發生融合和退化，保護染色體結構(chromosome structure )的穩定性和遺傳完整性(geneticintegrity)。端粒（Telomere）是一個人衰老速度最重要、最準確的指標。它的初始長度是由遺傳和環境因素決定的，並且會隨著時間的推移而減少。研究表明，端粒（Telomere）長度與DNA修復(DNA repair)、衰老(aging)、細胞凋亡(apoptosis)和腫瘤發生(tumorigenesis)密切相關。因此，準確、可重複地測量端粒長度對研究人員來說尤為重要。本產品主要採用相對定量qPCR直接比較樣本平均端粒長度(telomere length)，即以端粒重複序列（telomererepeatsequence, Tel）與基因組單拷貝基因(genome single copy gene. SCR）拷貝數（T/S）的拷貝數比（T/S）作為端粒相對長度(telomere relative length)。單拷貝基因引物 (Single-copy gene primers, SCR) 特異性識別和擴增人類染色體上( humanchromosome)的 78bp 長區域。試劑盒中的引物組已通過檢測，確保：（1）高效可靠的定量； (2)非非特異性擴增。每組引物均經過擴增曲線效率驗證( amplification curve efficiency)（E>98%，R 2>0.99)，熔解曲線分析和凝膠電泳驗證。

Reaction System

A reaction system as described below was established. To perform multiple reactions, prepareapremixofthe common components, add a suitable volume to each tube or well, and then add a special reactioncomponent (eg, template).

1. It is recommended to use a 20μL system to ensure the validity and repeatability of the amplificationofthe gene of interest.

2. Cover or seal the reaction tube/PCR plate and mix gently. It can be centrifuged slightly toensurethatallcomponents are at the bottom of the tube.

3. Place the reaction system in a real-time PCR instrument, collect data and analyze the results. Setupyour PCR instrument as shown in the table below. Optimum temperature The incubation time can be determined by the specific situation.

Note: The main factors that determine the optimal annealing temperature are primer lengthandprimerbase composition. According to the characteristics of the kit’s telomere and SCR primer set, werecommend setting the annealing temperature to 55°C.
