谷氨醯胺合成酶(Glutamine Synthetase Activity, GS)主要存在於植物中，是體內同化氨(assimilation of ammonia)的關鍵酶之一。(Glutamine Synthetase Activity, GS)能催化銨離子和谷氨酸合成谷氨醯胺Gln( glutamine Gln)，不僅可以防止過量的生物毒性銨離子(biological toxic ammonium ions)，而且谷氨醯胺Gln( glutamine Gln)主要以氨的形式儲存和運輸。在ATP和Mg2+存在下，(Glutamine Synthetase Activity, GS)能催化銨離子和谷氨酸合成谷氨醯胺Gln( glutamine Gln)；谷氨醯胺Gln( glutamine Gln)進一步轉化為γ-谷氨醯異羥肟酸(gamma-glutamyl hydroxamic acid)，在酸性條件下形成紅色鐵絡合物；該絡合物在 540nm 處有最大吸收峰。

1. For cell and bacteria samples

Collect cell or bacteria into centrifuge tube, discard the supernatant after centrifugation, add 1 ml Assay buffer for 5 × 10⁶ cell or bacteria, sonicate (with power 20%, sonication 3s, intervation 10s, repeat 30 times); centrifuged at 8000g 4 °C for 10 minutes, take the supernatant into a new centrifuge tube and keep it on ice for detection.

2. For tissue samples Weigh out 0.1 g tissue, homogenize with 1 ml Assay buffer on ice, centrifuged at 8000g 4 °C for 10 minutes, take the supernatant into a new centrifuge tube and keep it on ice for detection.

3. For liquid samples Detect directly.
