泛素 C 末端水解酶 1 (Ubiquitin C-terminal Hydrolase 1 , UCH-L1) 是一種去泛素化酶 (deubiquitinating enzyme, DUB)，主要在神經元組織( neuronal tissue)中表達，在某些腦組織(brain tissues)中佔總蛋白質的 1-2%。 UCH-L1 是一種半胱氨酸蛋白酶(cysteine protease)，能夠切割泛素分子(ubiquitin molecule)的羧基端( carboxyl end )與泛素修飾蛋白上的賴氨酸殘基之間的異肽鍵(isopeptide bond )。由於泛素化(ubiquitination)用於這些修飾蛋白的定位和降解，因此去泛素化酶的正常功能對於細胞活力和完整性至關重要。 UCH-L1 的失調與帕金森病(Parkinson’s )和阿爾茨海默病(Alzheimer’s diseases)等神經系統疾病以及癌症侵襲(cancer invasion)的病理生理學有關。通過鑑定具有高選擇性的抑制劑可以促進對這些過程的進一步理解。已知有 400 多種 DUB 酶，該靶標提供了比蛋白酶體系統高得多的選擇性。 BioVision/ABCAM 的 UCH-L1 抑制劑篩選試劑盒(BioVision’s UCH-L1 Inhibitor Screening Kit) 利用活性 UCH-L1 去泛素化酶，切割合成蛋白底物以釋放游離螢光團的能力，可以使用螢光酶標儀輕鬆量化（Ex/Em = 350/440 nm）。小分子抑制劑可以減少或消除這種活性。因此，該抑制劑篩選試劑盒可以快速可靠地確定各種化合物對 UCH-L1 去泛素化酶的抑制作用，並應用於篩選新型抑制劑。
