Thiazolyl Blue (MTT) is a cell-permeable and positively charged tetrazolium dye that is used to detect reductive metabolism in cells. Thiazolyl Blue is taken up by cells through the plasma membrane and then reduced to formazan by intracellular NAD (P) H-oxidoreductases. Thiazolyl Blue is frequently used in colorimetric assays to measure cell proliferation, cytotoxicity, and apoptosis. Storage: protect from light.

MTT工作液的配製 (每種細胞條件不同)
用 PBS 溶解 MTT 以獲得 5 mg/mL 的 MTT。
細胞增殖檢測（96孔板）
1. 接種細胞：用含10%FBS的培養基製備單細胞懸液，接種於96孔板中，每孔1000-10000個細胞，每孔體積100uL。
2. 培養細胞：37℃。 5% CO2，孵育 24-72 小時。
3. 每孔加入10 μL MTT，孵育4 h，棄上清，懸浮細胞需先離心。
4. 加入100 μL DMSO，震盪10 min使晶體完全溶解
5. 用吸光度板讀數器在 OD = 562 nm 處監測吸光度增加。
Preparation:
It is soluble in water (10 mg/ml), ethanol (20 mg/ml) and 2-methoxyethanol (20mg/ml).
It is also soluble in buffered salt solutions and culture media (5 mg/ml).
Storage/Stability:
MTT is light sensitive.
Reconstituted MTT solution is stable for at least 6 months when stored frozen (-20 °C).
Storage of a reconstituted MTT solution at 2 – 8 °C for more than 2 weeks may cause decomposition and yield erroneous results.
