H2DCFDA (DCFH-DA) is a cell-permeable probe used to detect intracellular reactive oxygen species (ROS) (Ex/Em=488/525 nm)

H₂DCFDA (2′,7′-Dichlorodihydrofluorescein diacetate) is a useful fluorogenic reagent to detect reactive oxygen intermediates in cells. On oxidation, H₂DCFDA becomes the highly green fluorescent 2′,7′-dichlorofluorescein.

建議使用(依不同細胞需最佳化調整) 

1. H2DCFDA 溶於 DMSO 得到 10 mM 儲備液，使用前進一步稀釋。
2. 細胞與 5 µM H2DCFDA 溶液在 PBS 中於 37°C 避光孵育 30 分鐘，然後用 0.05% 胰蛋白酶-EDTA 溶液收集細胞，懸浮在新鮮培養基中，立即用流式細胞儀分析。
3. 如果需要，與 H2DCFDA 探針一起使用螢光素染料 DCFDA 的 ROS 不敏感修飾作為陽性對照。染色程序與 H2DCFDA 相同。
