巨噬細胞(Macrophages)是從循環的骨髓來源(circulating bone marrow-derived )的單核細胞(monocytes)分化而來的細胞。在骨髓(bone marrow )中以及隨後在血液和組織中，單核細胞(monocytes)經歷一系列成熟(maturation)，最終形成形態和功能成熟的組織巨噬細胞(tissue macrophages)。巨噬細胞的主要功能是清除細胞碎片和消滅入侵的病原體。它們吞噬入侵的微生物(microorganisms)，並清除(scavenge)死亡(dead)、受損的細胞(damaged cells )和細胞碎片( cellular debris)。巨噬細胞(Macrophages)可以通過流式細胞術( flow cytometry)或免疫組織化學染色( immunohistochemical stainin)通過幾種細胞表面蛋白(cell surface proteins)的特異性表達來鑑定，包括 CD14、CD11b、F4/80（小鼠）/EMR1（人）、MAC-1/MAC-3 和 CD68。

巨噬細胞培養基 (Macrophage Medium, MaM) 與巨噬細胞生長添加劑 (Macrophage Growth Supplement, MaGS, Cat #1972) 和 25 ml 胎牛血清 (FBS) 一起使用時，是專為正常巨噬細胞(normal macrophages)體外最佳生長而設計的完全培養基。它是一種無菌(sterile)液體培養基，含有必需(essential )和非必需氨基酸(non-essential amino acids)、維生素(vitamins)、有機(organic )和無機化合物(inorganic compounds)、激素(hormones)、生長因子(growth factors)、微量礦物質(trace minerals)和低濃度胎牛血清 (5%)。該培養基是 HEPES 和碳酸氫鹽緩衝液，在 5% CO2/95% 空氣的培養箱中平衡時的 pH 值為 7.4。該培養基經過配製（定量和定性）以提供最佳平衡的營養環境，選擇性地支持體外正常巨噬細胞的生長。
