DNase I (脫氧核糖核酸酶I, 即Deoxyribonuclease I)（IV 型）是一種從牛胰腺(bovine pancreas)中分離出來的酶。可消化單鏈或雙鏈DNA的脫氧核糖核酸內切酶， 它識別並切割磷酸二酯鍵，產生5′ 端為磷酸基團，3′ 端為羥基的單脫氧核苷酸或單鏈或雙鏈的寡脫氧核苷酸。生物活性≥ 2000 Kunitz 單位/mg 蛋白質在 pH 5.0 和 25 °C 下，使用 I 型或 III 型 DNA 作為底物，一個 Kunitz 單位將產生每分鐘每毫升 0.001的 A _260變化。
