蛋白質保存型式
蛋白質溶於20mM Tris, 150mM NaCl, pH8.0, containing 1mM EDTA, 1mM DTT, 0.01% SKL, 5% Trehalose and Proclin300.為冷凍乾燥包裝。使用前Reconstitute in 20mM Tris, 150mM NaCl (pH8.0) to a concentration of 0.1-1.0 mg/mL. Do not vortex.避免vortex. 短期保存Store at 2-8°C for one month.分裝-80°C長期12 個月。請避免反覆冷凍解凍。
纖連蛋白 (Fibronectin , FN) 是細胞外基質(extracellular matrix)的一種高分子量 (~440kDa) 糖蛋白(glycoprotein)，可與稱為整合素(integrins)的跨膜受體蛋白(membrane-spanning receptor proteins)結合。纖連蛋白(Fibronectins, FN)結合細胞表面(cell surfaces)和各種化合物(various compounds)，包括膠原蛋白(collagen)、纖維蛋白(fibrin)、肝素(heparin)、DNA 和肌動蛋白。纖連蛋白(Fibronectins, FN)具有多種功能。例如，它參與細胞粘附( cell adhesion)、細胞運動(cell motility)、調理作用(opsonization)、傷口癒合(wound healing)、細胞形態維持(maintenance of cell shape)等。此外，結締組織生長因子 (CTGF) 已被確定為 纖連蛋白(Fibronectins, FN)的相互作用因子，因此進行結合 ELISA 測定以檢測重組人纖連蛋白(Fibronectins, FN) 和重組人 CTGF 的相互作用。簡而言之，將 纖連蛋白(Fibronectins, FN) 在含有 0.01% BSA (pH 7.4) 的 PBS 中連續稀釋。然後將 100uL 的重複樣品轉移到 CTGF 包被的微量滴定孔中，並在 37℃ 下孵育 2 小時。用PBST洗滌孔並用抗纖連蛋白(Fibronectins, FN)pAb孵育1小時，然後吸出並洗滌3次。用HRP標記的二抗孵育後，將孔吸出並洗滌3次。加入底物溶液，37℃孵育15-25分鐘。最後，向孔中加入 50µL 終止液，立即在 450nm 處讀數。 FN與CTGF的結合活性如圖1所示，這種作用呈劑量依賴性。
