核酸外切酶III(Exonuclease III),大腸桿菌從切口DNA(nicked DNA),鈍端DNA(blunt end DNA),3′-凹陷末端(3′-recessed ends)或少於4個鹼基的3′-突出端沿3’→5’方向消化雙鏈體DNA,並生成核苷5′-磷酸。 DNA降解以均勻的速度進行,並在相反的鏈上產生ssDNA片段。在確定的反應條件下,反應可產生可預測和可再現結果。需要優化條件,例如溫度(temperature),離子強度(ionic strength),模板DNA(template DNA)序列和核酸外切酶III與DNA的比例,以適合特定應用以實現所需的切除率。由於固有的3′-磷酸酶活性(3′-phosphatase activity),外切核酸酶III(Exonuclease III),大腸桿菌也能夠從3′-磷酸末端降解DNA。該酶還具有嘌呤DNA核酸內切酶活性以及RNase H活性。

| Description | Exonuclease III, E.coli digests duplex DNA in the 3′→5′ direction from nicked DNA, blunt end DNA, 3′-recessed ends, or 3′-overhangs of less than four bases, and yields nucleoside 5′-phosphates. The DNA degradation proceeds at a uniform rate and produces stretches of ssDNA on the opposite strand. Under defined reaction conditions, the reaction can yield predictable and reproducible digestion results. Conditions such as temperature, ionic strength, template DNA sequence and the Exonuclease III to DNA ratio need to be optimized to suit specific applications to achieve the desired excision rate. Exonuclease III, E.coli is also capable of degrading DNA from 3′-phosphate ends due to intrinsic 3′-phosphatase activity. The enzyme also has apurinic DNA endonuclease activity as well as RNase H activity. |
