抗體介紹: 此抗體特異性針對PD-1H抗原。The antibody has been labelled with biotin and used to stain formalin-fixed tissues from wild-type C57BL/6 mice at 6 weeks of age. This showed specific staining for PD-1H in the T cell zone and marginal zone. The antibody stains P815 cells transfected with full-length PD-1H plasmid, but not control plasmid. Antibody specificity has been confirmed by ELISA. A single 200-μg dose of MH5A prevented aGVHD in mouse models, caused a reduction in accumulation and expansion of CD8+ and CD4+ T cells in spleens and livers and greatly reduced numbers of inflitrating T cells in all aGVHD target tissues examined. Few mAbs targeting a single cosignalling molecule have such a potent preventative effect (Flies D et al, 2011).。可應用於therapeutic, Block, ELISA, IHC等實驗。Species Reactivity: Mouse

Immunofluorescence staining of fixed mouse splenocytes with anti-PD-1H antibody MH5A (Ab01016) Immunofluorescence analysis of paraformaldehyde fixed mouse splenocytes permeabilized with 0.15% Triton and stained with the chimeric mouse IgG1 version of MH5A (Ab01016-23.0) at 10 µg/ml for 1h followed by Alexa Fluor® 488 secondary antibody (2 µg/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab01016-23.0, DAPI, merged channels and an isotype control. The isotype control was stained with an anti-unknown specificity antibody (Ab178-23.0) followed by Alexa Fluor® 488 secondary antibody.

ELISA of anti-VISTA antibody on VISTA-Fc fusion protein. Binding curve of the rabbit chimeric version of the anti-VISTA antibody MH5A (Ab01016-23.0; blue line) to an ELISA plate coated with mouse VISTA-Fc fusion protein (Pr00164-1.9) at a concentration of 5 µg/ml. A 3-fold serial dilution from 10000 to 1.7 ng/ml was performed using Ab01016-23.0. For signal detection, a 1:4000 dilution of anti-rabbit IgG1 HRP (BioRad) antibody was used.
